Good but need to be improved
Application or Technique: DNA and protein transfer

IBlot is good for quickly transfer your protein or DNA into a blot membrane. It can save your 30 to 50 min for protein transfer, save you 20min at least for DNA transfer and transfer more efficient than capillary transfer. However, the hinge is easy to be broken. Besides that, you have to buy the transfer stacks, that is not cheap...

The iBlot is a great design, my only issue with it is the amount of waste, if this could be reduced a little with re-usable sponges etc it would be 5 stars all around

This is the only device I have ever used to do transfers, and it was one of the first pieces of equipment that I learned to use in the lab. It is very simple and easy to use, and it is effective. According to what I have heard about the old way of doing this, this device seems excellent. Setting P3 for 8 minutes is perfect for everything we have run into so far. BioRad TGX pre-cast gels need to be trimmed some at the sides to not hang out of the sides of the transfer stack.

The iBlot system is excellent for protein transfer and is my lab's current preferred method. It is simple to use, provides consistent data, and the time savings is well worth the investment in the system when working with undergraduate students that have little time to spare. My only complaint is that it is an expensive system to use for an academic laboratory with many students and somewhat low funds.

A western blotter's dream!
Application or Technique: Immunoblotting

The iBlot is a western blotter's dream! 7 min transfers, all with premade buffers and membrane sandwiches. High molecular weight proteins sometimes do not transfer completely so you may have to adjust the transfer time based on the size of your target protein. But, overall, it is a fantastic product.

Overall the instrument works very well with the majority of protins i am trying to transfer. It is very easy and convenient to use. In some caase proteins do not transfer completely compared to wet transfer but ease of use still makes it the preferred step.

Midi gels and the iBlot
Application or Technique: WB
Institution: Boston University
Hi, I have used the iBlot system with Midi gels for two years now. I think the system works well and I find that I get a comparable transfer of proteins to the membrane compared to traditional blotting techniques. But the system is a little challenging at first i.e. getting used to handling the thin midi gels and setting the blotting “sandwich” without air bubbles in-between.

The iBlot instrument is a teriffic time saver without sacrificing performance. Using the instrument saves the time required to set up and run a standard Western transfer and also has a green component to it as it does not generate methanol containing waste.

The iBlot is much easier to teach novice students how to use than traditions buffer transfer systems. The transfer is fast and works with homemade gels. We found that cutting the stack to fit the gel helped with transfer issues - also adjusting the time of transfer from 7 mins to 8 mins enabled a better transfer of our higher molecular weight proteins. The down side is, for the price paid for the device and the stacks, the manufacturer could have invested more time into a more sturdy latch.
